Conventional means for delivering active agents are often severely limited by biological, chemical, and physical barriers. Typically, these barriers are imposed by the environment through which delivery occurs, the environment of the target for delivery, and/or the target itself. Biologically or chemically active agents are particularly vulnerable to such barriers.
In the delivery to animals of biologically active or chemically active pharmacological and therapeutic agents, barriers are imposed by the body. Examples of physical barriers are the skin, lipid bi-layers and various organ membranes that are relatively impermeable to certain active agents but must be traversed before reaching a target, such as the circulatory system. Chemical barriers include, but are not limited to, pH variations in the gastrointestinal (GI) tract and degrading enzymes.
These barriers are of particular significance in the delivery of orally administered pharmacological agents. For example, orally administered pharmacological agents are subject to digestion and absorption by the GI tract and the resulting chemical, biological and physical barriers are clearly evident. For example, shortly after administration, only about 0.1 to about 1% of an orally administered pharmacological agent reaches the systemic circulation. The agent is exposed first to acidic pH in the stomach and first to biocatalytic enzymes in the small intestine.
In addition, the effective dose of an orally administered agent may be restricted by the degree of absorption, distribution throughout the body, metabolism or excretion. For example, the blood-brain barrier limits delivery of many therapeutic agents designed for CNS delivery. The reduction in efficacy due to restrictions in drug delivery may be significant as demonstrated by a reduction in therapeutic benefit.
As another example, the use of biocompatible polymer matrices for the delivery of pharmacological agents as well as other active agents is attracting increased interest. One type of biocompatible polymer being used is poly(ethylene glycol) (“PEG”). PEG is a polymer having the formula H(OCH2CH2)nOH where n is often in the range of about 10 to about 100 or more. PEG is widely used in biomedical research to modify the biodistribution of drugs. The focus of PEG research has been to prolong the residence time of drugs or drug-like agents in the body by altering their biodistribution. It has been discovered that the polymer PEG has unique characteristics that are beneficial in the delivery of drugs. For example, PEG is often soluble in water and is non-toxic.
